This invention relates to the field of molecular diagnostics and nucleic acid detection. In particular, it concerns an isothermal assay platform for the multiplex detection of circulating microRNAs (miRNAs) in small-volume blood samples. The platform is useful for early detection, screening, or monitoring of diseases such as cancer and other conditions.

Background: 
Small noncoding RNAs, known as microRNAs (miRNAs), control the expression of genes that code for proteins by either cleaving RNA transcripts or suppressing translation. Numerous types of malignancies have been shown to have abnormal miRNA expression. Thus, miRNA-expression patterns can be used as biomarkers for cancer prediction, diagnosis, and therapy response. The distinctive features of miRNA, such as its short size, low cellular abundance, and high homology, make it difficult to build precise miRNA detection techniques, despite the efforts of many researchers. This invention provides an ultrasensitive and specific approach compared to traditional miRNA detection method.
