This invention relates to an ultrasound-transparent organ-on-chip platform and a method to study and monitor acoustic wave interaction with gas bubbles and disruption of cellular barriers. The invention can be used for drug screening for ultrasound gas bubble disruption of the blood-brain-barrier (BBB) for brain drug delivery and can be used as a product for research use.

Background: 
Ultrasound-based techniques can transiently open cellular barriers to enable drug delivery, but the underlying bubble-cell-acoustic interactions are complex and are difficult to observe directly in vivo. Unlike conventional in vitro assays, this platform integrates ultrasound compatibility, optical transparency, and physiologically relevant microfluidics, allowing real-time visualization of gas bubble dynamics while simultaneously measuring blood-brain barrier function under flow.
